反相高效液相色谱法直接检测芳香族氨基酸

摘要:
用反相高效液相色谱-变波长紫外检测法直接测定了芳香族氨基酸.流动相为磷酸盐缓冲溶液(pH6)和甲醇的混合溶液,其体积配比为70︰30,流速为0.6 ml / min,变波长紫外检测,10 min内完成.酪氨酸、苯丙氨酸、色氨酸的线性范围分别为50~500μg·mL-1、200~20000μg·mL-1、20~2000μg·mL-1,三者的检测限分别为10μg·mL-1、150μg·mL-1、5μg·mL-1.

Abstract:
A method for determination of aromatic amino acids by RP-HPLC with UV detection at changeable wavelength was developed . Chromatographic conditions were as follows: mobile phase of phosphate buffer (pH6.0) and methanol in a ratio of 70: 30; flow rate of 0.6 ml/min. Samples were finally analyzed in 10min. Linear range of tyrosine, phenylalanine and tryptophane was 50-500μg·mL -1 , 200-20000μg·mL -1 and 20-2000μg·mL -1 . Their detection limit was 10μg·mL -1 、150μg·mL -1 and 5μg·mL -1 .
